The drug Ganaton belongs to the pharmacological group of drugs used for functional disorders of the gastrointestinal tract.

Indications Ganatone

The drug Ganaton is prescribed to activate the gastrointestinal tract motility in gastroesophageal reflux disease (GERD), chronic gastritis, stomach and intestinal spasms. It can be used for dyspeptic symptoms (not associated with gastric ulcer and duodenal ulcer); gastralgia, nausea, vomiting, heartburn, flatulence, decreased or loss of appetite.

Release form

Film-coated tablets of 50 mg (in blister pack).

Other trade names: Itopride hydrochloride, Itopride, Itomed, Primer.

Pharmacodynamics

The active substance of the drug Ganaton is the prokinetic itopride hydrochloride - a blocker (antagonist) of dopamine receptors (D2) of the gastrointestinal tract. By affecting D2 receptors, the drug inhibits the functions of the catalyst of acetylcholine hydrolysis - the enzyme acetylcholinesterase.

This increases the intracellular level of acetylcholine in the nerve receptors of the gastrointestinal tract and provides energy to the smooth muscle cells of the digestive organs (due to the unimpeded transformation of ATP into cAMP). As a result, the tone of the muscle fibers of the gastrointestinal tract increases, and the motility of the digestive tract is activated.

Pharmacokinetics

Once in the gastrointestinal tract, up to 90% of itopride hydrochloride is absorbed through the intestinal walls into the bloodstream; the highest level of the drug in the blood plasma is achieved 40-45 minutes after its administration.

Ganaton is broken down by liver enzymes, there is no cumulative effect. Metabolites are eliminated in the urine with a half-life of about 6 hours.

Dosing and administration

Ganaton tablets are taken orally (whole, one hour before meals) - one tablet (50 mg) three times a day. The maximum daily dose is 150 mg. The standard course of treatment is 14-21 days.

Use Ganatone during pregnancy

Since the safety of Ganaton for pregnant women has not been studied, its use during pregnancy is not recommended.

Contraindications

Contraindications for Ganaton include: hypersensitivity to itopride hydrochloride, gastric bleeding, intestinal stenosis, lactation in women, age under 16 years.

Side effects Ganatone

The use of Ganaton may cause side effects such as skin allergic reactions; diarrhea, constipation, abdominal pain, increased dry mouth; headache, sleep disturbances; galactorrhea or gynecomastia (due to an increase in the level of the hormone prolactin in the blood); urinary retention (with a history of prostate hypertrophy).

Overdose

Overdose symptoms are not described in the official instructions. In case of overdose, gastric lavage should be performed.

Interactions with other drugs

Ganaton should not be taken simultaneously with anticholinergic drugs, as this reduces its therapeutic effect.

It is permissible to use this drug simultaneously with medications for the treatment of gastric ulcer.

Store at room temperature, protected from light.
